A T-DNA construct containing a promoter less gus reporter was introduced into the genome of Arabidopsis plants using Agrobacterium- mediated transformation. We have produced a collection of T-DNA tagged lines using the promoter-less gus vector pdgusBin19. A part of this collection was screened for identification of lines with specific gus expression patterns. Here we report identification of two lines with an interesting expression of gus in anthers and seeds. Both lines harboured complex T-DNA inserts. In the first line (MS) integration of the T-DNA insert caused a male sterility phenotype. gus expression is developmentally regulated in the anther and flower base. In the second line (ESL) expression of gus was seen in the anther and seed endosperm. For both lines attempts were done to isolate the T-DNA flanking regions. The gus and DNA analysis for MS and ESL will be discussed in detail.
