Abs # 137: Analysis of expressed sequence tags (ESTs) induced by oxidative stress from hot pepper

For the massive screening of the genes induced by oxidative stress, we constructed a cDNA library from hot pepper (Capsicum annuum L. cv. Nockwang) leaves treated with paraquat. From this cDNA library, 1589 cDNA clones were sequenced from their 5กฏ ends. The sequences were clustered into 1252 non-redundant groups comprised of 152 non-redundant clusters and 1100 singletons. Similarity search results against BLASTX identified 1005 (80.27%) clones for known proteins, 197 (15.74%) clones for unknown and 50 (3.99%) clones for no hit. Oxidative stress-related clones such as ascorbate peroxidase, catalase, osmotin precursor were detected from the ESTs. 789 ESTs were grouped into several categories such as metabolism, protein synthesis, cellular communication / signal transduction, cell rescue and defense. However, the other 463 ESTs showed no match with MIPS database. We will perform cDNA microarray analysis to isolate cDNA clones related to oxidative stress and to study their functional roles.
