Abs # 905: An efficient system for rapid generation of low copy transgenic plants using the maize inbred H99

A high efficiency transformation system was developed for the maize inbred H99. This system is based on bombardment of immature embryos or callus with plasmid DNA encoding NPTII as the selectable marker, and selection of transformants using the antibiotic paromomycin. Using this system, transformation frequencies exceeding 50% (events to soil/embryos or callus pieces bombarded) were observed. Precipitating low concentrations of DNA onto gold particles for bombardment was shown to enhance the recovery of low- and single-copy transgenic plants. High frequency recovery of transformants was demonstrated using as little as three weeks of selection.
