Abs # 935: Physical mapping and contig assembly using BAC clones in Chinese cabbage

A physical map used as effective means that can immediately access for any genome will make it possible to select clones for sequencing that would ensure comprehensive coverage of the genome. To conduct physical mapping of Chinese cabbage, each BAC clones of Chinese cabbage were digested with HindĽ˛, and separated on agarose gels. restriction fragments of each clones were called and normalized by the program IMAGE. Based on this results, contigs were created on the program FPC. In order of physical mapping of Chinese cabbage, we analyzed 18,189 BAC clones of Chinese cabbage (Feb. 2003). The results generated the 884 contigs which were contained 2,643 clones and 11,909 singletons based on tolerance value of 7 and cutoff value of 10-12. For constructing the contig assembly for whole genome of Chinese cabbage, we will apply to fingerprint 56,592 clones (HindĽ˛ BAC library) and be expected to construct BamHI BAC library for complete contig assembly.
