Proteomic approaches using two-dimensional polyacrylamide gel electrophoresis ( 2D-PAGE ) were carried out to identify the differentially expressed proteins from rice suspension-cultured cell and leaf responsive to rice blast fungus, M. grisea. Proteins were extracted and PEG fractionated from suspension calli and leaf over time course of 24 and 48hr after rice blast fungus inoculation. Twelve and ten protein spots were induced in the suspension-cultured cell and leaf inoculated with rice blast fungus, respectively. These proteins were analyzed by N-terminal or internal protein sequencer and mass spectrometry. The salt induced proteins (SalT), pathogen-related protein ( OsPR-10 ), isoflavone like reductase (OsIRL), b-glucosidase and novel protein were induced in the rice suspension cell. Two spots showed homology to putative receptor-like protein kinase having DUF 26 domain were induced in the inoculated leaf. This domain has no known function. Two b-1,3-glucanases (b-Glu1, -Glu2), pathogenesis-related thaumatin-like protein (TLP), and peroxidase (POX 22.3) were induced in the leaf. Six spots showed homology to probenazole-inducible protein (PBZ) were identified in the suspension-cultured cell and whole plant leaf. The expression level of these induced proteins was further confirmed by Northern/ Western blots and immunohistochemistry analysis.
