To dissect the signal transduction of ABA in plants, we studied the expression of the ABA-responsive rd29B gene in Arabidopsis plants, and isolated Arabidopsis mutants altered in expression of the rd29B gene. We compared the expression of the rd29B gene and those of genes encoding transcription factors such as AREB1 and ABI5 by northern analysis. We also compared the expression of around 7,000 Arabidopsis genes under ABA treatments during germination and that in mature plants using a full-length cDNA microarray. The results indicated that ABA-responsive gene expression during germination is not same to that in mature plants. We introduced a chimeric gene construct consisting of the luciferase under the control of the rd29B promoter ( rd29B - LUC ) into Arabidopsis plants. Activation tagged lines of Arabidopsis containing rd29B - LUC were obtained by transformation; mediated by Agrobacterium carrying the vectors pPCVICEn4HPT or pSKI015. We also obtained mutant seeds using EMS treatment. We screened mutants altered in ABA-response of the rd29B - LUC gene during germination or under stress conditions in mature plants. Some mutants altered in the regulation of the rd29B - LUC gene were identified. Most of the isolated mutants were altered in ABA-responsive expression of the gene during germination and in mature plants. Some of them were altered in ABA-responsive expression of the gene during germination or in mature plants. Analysis of the mutants is in progress.
