Abs # 1419: Detection of an Allelic Series in Soybean and Wheat through TILLING

We are using a molecular approach to improve crops by identifying induced variation in genomes. Through the application of TILLING (Targeting Induced Local Lesions in Genomes), we have identified multiple alleles of genes of interest and developed molecular markers for crop improvement. This method involves inducing variation by treating seeds with EMS, preparing DNA from M2 plants, pooling samples, PCR amplifying the genes of interest, and detecting novel alleles by either a Cel 1 method or by Temperature Gradient Capillary Electrophoresis (TGCE). The Cel 1 method involves the enzymatic cleavage of dye-labeled heteroduplexed DNA followed by PAGE. TGCE involves detection of heteroduplex DNA separated from homoduplex DNA by its altered migration through a capillary over which a temperature gradient has been applied (SpectruMedix). We have applied TILLING to tomato, zebrafish, corn, rice, lettuce, peanut, wheat, and soybean and will present data from soybean and wheat.
