Abs # 1426: Construction and profile analysis of a cDNA library from young fruits of Satsuma mandarin

A cDNA library of ¡®Nichin 1 Gou¡¯ Satsuma mandarin (C. unshiu) was constructed from young fruits and the profile of expressed genes was analyzed. The total mRNA was isolated from whole fruits including peel and pulp 30days after flowering. The full-length cDNA library was prepared from mature mRNAs with both of 5¡¯Cap and polyA tail and the 2350 cDNA clones were partially sequenced. The result of cluster analysis of the cDNA library with CAP3 sequence assembly program showed the 662 clones to be singletons and the other 1688 clones to form 145 contigs. Therefore the number of unigene with unique sequence was 807. According to the homology search with BLASTX against NCBI database, 377 unigenes are highly homologous to known genes and 430 unigenes are unknown. Among the unknown 430 unigenes, the 159 genes had homology with some peptides in NCBI database, however the functions of the peptides are unknown. The other 271 unigenes had no homology with any other peptide in NCBI database, therefore these seemed to be novel genes expressed in young fruits of Satsuma mandarin. The most abundant class of genes expressed in young fruits of Satsuma mandarin was chitinase. The total number of cDNA clones homologous to chitinase genes was 170 and these were categorized into 4 types of chitinase. Also 99 cDNA clones were homologous to 3 types of lectin genes, and 77 cDNA clones were homologous to 2 types of miraculin genes. These three classes of highly expressed genes, chitinases, lectins and muraculins seemed to play important roles in early developmental stage of the citrus fruits.
