Abs # M1004: The two cell-cycle regulated set-domain proteins ATRX5 and ATR6 interact with PCNA

The Proliferative Nuclear antigen (PCNA), recognized as a DNA polymerase processivity factor, has direct interactions with various proteins involved in the important genetic information processes in Eukarya.. PCNA is involved in DNA repair, maintenance of hetechromatic regions throughout replication, cell cycle regulation and Programmed cell death. Identification of PCNA partners is therefore necessary to understand these processses. Two Arabidopsis SET-domain proteins ATRX5 and ATRX6 were isolated from a two hybrid screen using PCNA.as a bait. A deletion of the PIP-box located in ATRX5 exon 2 prevents its interaction with PCNA. This ATRX5 spliced form is present all along the plant development. Although ATRX5 showed a double localisation in plastids and in nuclei, ATRX6 localisation is restricted to nuclei.ATXR6, up-regulated in S-phase is a direct target to E2F transcription factor. Over-expression of the two ATRX proteins causes male sterility due to the degeneration of defined cell type, suggesting they could be involved in the control of cell death pathway.
